Hello,

our Provider STRATO told us that in future there will be the need of authentication for sending mail rather than authentication via prior pop3 connect.

How can i configure this in mercury/32 v401b? 

I made an account at strato smtpauth@domain.de and wrote the data in the fields in mercuryC smtp relay client under "credentials for smtp...." etc, fields "Username" and "Password" .

Is that enough for getting it to work?

You do not, it does not have that capability.  You need to upgrade to Mercury/32 v4.62 to do this.  This version of MercuryC has the capability to do both SSL/TLS and ESMTP Authentication.

Yes, assuming that your provider uses one of the standard authentication methods.  David Harris has implemented ESMTP AUTH CRAM-MD5 for WinPMail, Mercury and Mercury/32.  They do a SMTP authentication in accordance with RFCs 2554 and 2195.  Mercury and Mercury/32 also do the LOGIN and the strange MS AUTH=LOGIN.
