This is a really old discussion to reactivate and you have not provided any information specific to your situation. If it is the same as the OP (drive letter change on an external drive) the I think the easiest solution is to assign the previous drive letter to that drive via Computer Management.
If you are unable to do that, have you read through the thread of the post linked to by Michael? It does a good job of explaining how to configure Pegasus Mail to use mailboxes at a specific location. One thing to be aware of is that the utility "pconfig.exe" referenced in the thread is a 16 bit DOS app that won't run on current machines without a DOS emulator. You would use pconfig32.exe instead (included with Pegasus Mail v4.71+).
